| I. Background | |
| T 細胞免疫球蛋白和粘蛋白結構域 3（TIM3 ）是一種含有免疫球蛋白和粘蛋白結構域 的細胞表面分子，最初被發(fā)現(xiàn)為 IFN-γ產生 CD4 T 輔助蛋白 1 （Th1） 和 CD8 T 細胞毒性 1 （Tc1） 細胞的細胞表面標記物，是一種關鍵的免疫檢查點受體，主要在活化的 CD8+T 細胞、耗竭性 T 細胞、Th1 細胞及先天免疫細胞（如樹突細胞、巨噬細胞）中表達。TIM 蛋白家族是具有相似結構的 I 型膜蛋白：可變 Ig 結構域（IgV）、不同長度的糖基化粘蛋 白結構域和單個跨膜結構域。除 TIM4 外，所有 TIM 分子都含有 C 端細胞質尾部，具有保 守的基于酪氨酸的信號轉導基序。 目前的 TIM3 信號傳導模型是，在 T 細胞激活時，TIM3 被募集到免疫突觸，其中 Bat3 與 TIM3 的細胞質尾部結合，并募集活性催化形式的淋巴細胞特異性蛋白酪氨酸激酶 （Lck）。然而，當 TIM3 與配體結合時，細胞質尾部中保守的酪氨酸殘基被磷酸化，導致 Bat3 的釋放，從而使 TIM3 發(fā)揮其抑制功能。 |
